Product Specific Information

Description:
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, a subset of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a crucial role in T cell development and activation. In T cells, CD4 associates with protein tyrosine kinase p56lck via its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Reported:
This antibody has been reported for use in flow cytometry, immunoprecipitation, and IHC of frozen sections. It is also used for isolation and depletion of CD4+ cells, and for blocking CD4-ligand binding and T cell activation.

Applications Tested:
Tested by flow cytometric analysis of mouse thymocytes and splenocytes. Recommended ≤0.125 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Optimize dilution for specific assays.

Endotoxin:
< 0.05 ng/µg antibody (LAL assay)

Target Information

CD4 is a co-receptor for MHC class II molecules and HIV receptor. It is primarily expressed on T helper cells, but also found on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 participates in T-cell activation through association with the TCR complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
