Reconstitution:

• Add 25 µL, 50 µL, or 0.2 mL double-distilled water (DDW), depending on sample size.
• Ships as lyophilized powder at room temperature.
• Upon arrival, store at -20°C.
• Reconstituted solution can be stored at 4°C for up to 1 week.
• For long-term storage, aliquot and store at -20°C.
• Avoid multiple freeze-thaw cycles.
• Centrifuge antibody preparations before use (10,000 × g, 5 min).

Target Information

TRPC6 forms a receptor-activated calcium channel in the cell membrane.
It is activated by diacylglycerol (DAG) and regulated through a phosphatidylinositol second messenger system, independent of protein kinase C and intracellular calcium levels.
Mutations in TRPC6 are associated with focal segmental glomerulosclerosis 2 (FSGS2).

TRPC6 belongs to the “canonical TRP” (TRPC) family, part of the mammalian transient receptor potential (TRP) superfamily.
Members of the TRPC3/6/7 subfamily are activated by DAG analogs, suggesting regulation via PLC-coupled receptors.
TRPC6 functions as a non-selective, store-independent, receptor-activated cation channel and is insensitive to IP3 activation.
It encodes endogenous DAG-activated receptor-operated cation channels in vivo.
