Target Information
Trpc6 forms a receptor-activated calcium channel in the cell membrane. It is activated by diacylglycerol (DAG) and functions independently of protein kinase C. TRPC6 plays a role in phosphatidylinositol-mediated signaling and calcium regulation. Mutations in the TRPC6 gene are associated with focal segmental glomerulosclerosis 2 (FSGS2).
TRPC6 belongs to the “canonical TRP” (TRPC) family, part of the mammalian transient receptor potential (TRP) superfamily. Members of this family are activated through PLC-coupled receptors and act as receptor-regulated cation channels. TRPC6 channels can be activated by DAG analogs, suggesting a PLC-coupled receptor mechanism.
When expressed in transfected cells, TRPC6 acts as a non-selective, store-independent, receptor-activated cation channel. It is DAG-activated, PKC-independent, and insensitive to IP3. Increasing evidence supports TRPC6 as an endogenous DAG-activated receptor-operated cation channel in vivo.
Diseases associated with TRPC6 include Glomerulosclerosis, Focal Segmental, 2 and Focal Segmental Glomerulosclerosis.
