Target Information

Tau is a neuronal microtubule-associated protein primarily located in axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus associates with neural plasma membrane components, functioning as a linker between cytoskeletal and membrane structures.

Tau localization determines axonal polarity within neurons. Short isoforms contribute to cytoskeletal plasticity, whereas longer isoforms aid stabilization. Hyper-phosphorylated Tau forms paired helical filaments (PHF), the main component of neurofibrillary tangles in Alzheimer’s disease (AD). This modification disrupts microtubule binding, leading to neuronal degeneration.

Tau phosphorylation is mediated by several kinases, including GSK-3β, PKA, cdk5, and casein kinase II. Hyper-phosphorylated Tau is implicated in Alzheimer’s disease, Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
