Target Information

The oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase subfamily, which includes EGFR, ERBB2, and ERBB3. ERBB4 binds and is activated by neuregulins and other ligands, inducing cellular responses such as mitogenesis and differentiation.
ErbB4 acts as a receptor for heregulin and mediates ligand-stimulated tyrosine phosphorylation, regulating key cellular processes including metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, apoptosis, and differentiation.
It is predominantly expressed in breast carcinoma cell lines (T47-D, MDA-MB-453, BT-474, H3396) and normal tissues such as skeletal muscle, heart, pituitary, brain, and cerebellum. Expression is low or absent in some breast tumor cell lines (MDA-MB-231, MDA-MB-157, MDA-MB-468, SKBR-3). Elevated ErbB4 levels in certain tumors suggest a role in malignancy.
