Target Information
Apoptosis Inducing Factor (AIF) causes chromatin condensation and DNA fragmentation. AIF was recently identified and cloned. Apoptosis is characterized by several morphological nuclear changes including chromatin condensation and nuclear fragmentation triggered by activation of caspase family members and other proteins.
AIF localizes in mitochondria and translocates to the nucleus upon induction of apoptosis, promoting release of apoptogenic proteins such as cytochrome c and caspase-9. It induces chromatin condensation and large-scale DNA fragmentation, hallmarks of apoptosis, both in isolated nuclei and in live cells.
AIF is highly conserved between human and mouse and widely expressed. Mutations in the AIF gene cause combined oxidative phosphorylation deficiency 6, resulting in severe mitochondrial encephalomyopathy. Alternative splicing generates multiple transcript variants; a related pseudogene exists on chromosome 10.
