Target Information

The second messengers cAMP and cGMP are key regulatory molecules involved in diverse signal transduction pathways. Their levels are controlled by synthesis via nucleotide cyclases and hydrolysis by cyclic nucleotide phosphodiesterases (PDEs).
PDEs form a superfamily of enzymes converting 3′,5′-cyclic nucleotides into nucleoside 5′-monophosphates. PDE6 acts as the effector enzyme in the G protein-mediated visual signal transduction cascade.

PDE6 consists of several subunits:

• Catalytic subunits: α′ (Cone), α (Rod), and β (Rod)
• Inhibitory subunit: γ
• δ subunit: function unknown but likely interacts with other proteins beyond the PDE6 family

The catalytic core comprises α′/α′ homodimers in cones and α/β heterodimers in rods.
Both catalytic and inhibitory subunits have C-terminal modifications (methylation, myristoylation, prenylation) essential for complex assembly and membrane association.
