Product Specific Information
This antibody was negatively preadsorbed using:
- A non-phosphopeptide corresponding to the phosphorylation site to remove reactivity with non-phosphorylated paxillin.
- A generic tyrosine-phosphorylated peptide to remove reactivity with phosphotyrosine irrespective of sequence.
The final antibody was affinity-purified using a paxillin-derived peptide phosphorylated at tyrosine 118.
Positive controls for Western blotting:
- Normal mouse mammary gland (NMuMG) cells
- NMuMG cells transfected with EGFP-tagged paxillin ± TGFβ
- Human epidermoid carcinoma cells (A431) ± EGF
Target Information
Paxillin is a focal adhesion protein and a substrate for several tyrosine kinases such as Src, FAK, and p210BRC/ABL. Its tyrosine phosphorylation is influenced by changes in cell-cell adhesion. Paxillin interacts with FAK and Crk via its SH2 domain, independently of the extracellular matrix. Although initially identified in fibroblasts, paxillin phosphorylation also plays a role in neurite extension during differentiation.
