Product Specific Information
Purified from rabbit serum by sequential epitope-specific chromatography. The antibody has been negatively preadsorbed using a non-phosphopeptide to remove reactivity with non-phosphorylated Tau. The final product is affinity purified using a Tau-derived peptide phosphorylated at threonine 205.
This antibody has been used in western blotting and immunohistochemistry. In western analysis, recombinant human Tau treated with GSK3β was used as a positive control.
Each vial contains sufficient material for approximately 10 mini-blots.
Target Information
Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus binds plasma membrane components, functioning as a linker protein.
Hyper-phosphorylated Tau forms paired helical filaments (PHF), the major component of neurofibrillary tangles in Alzheimer’s disease. Phosphorylation impairs microtubule binding, leading to neuronal degeneration.
Tau phosphorylation is mediated by multiple kinases, including GSK-3β, PKA, cdk5, and casein kinase II. Hyper-phosphorylated Tau is implicated in Alzheimer’s, Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
