Target Information
The E3 ubiquitin-protein ligase NEDD4 is a Ubl conjugation pathway protein containing one HECT (E6AP-type E3 ubiquitin-protein ligase) domain and four WW domains.
It participates in degradation of VEGFR-2/KDFR independently of ubiquitin-ligase activity and acts as a potential proto-oncogene by negatively regulating PTEN via ubiquitination.
NEDD4 is also involved in viral budding and regulation of epithelial Na⁺ channels, contributing to arterial hypertension.
Localized to the sarcolemmal region of muscle fibers, increased NEDD4 expression is associated with muscle unloading and denervation, as well as decreased Notch1 expression, functioning as a down-regulator of Notch-1 gene expression.
