Target Information
IRS-1 is a major substrate of the insulin receptor and is phosphorylated in response to stimulation by insulin, insulin-like growth factor 1 (IGF-1), and interleukin 4 (IL-4). It undergoes serine, threonine, and tyrosine phosphorylation across various tissues.
Casein kinase II mediates part of the insulin-stimulated serine/threonine phosphorylation of IRS-1 in vivo, with Thr502 and Ser99 identified as major phosphorylation sites. This phosphorylation may be a component of the intracellular insulin signaling cascade.
IRS-1 also contains three putative binding sites for 14-3-3 (Ser270, Ser374, and Ser641), with the Ser270 motif located in the phosphotyrosine binding domain responsible for interaction with the insulin receptor.
