Target Information
TRPC6은 세포막의 수용체 활성화 칼슘 채널을 형성합니다. 이 채널은 diacylglycerol(DAG)에 의해 활성화되며, phosphatidylinositol 2차 신호전달계에 의해 조절되는 것으로 알려져 있습니다. TRPC6 활성화는 protein kinase C(PKC)와 무관하며 낮은 세포내 칼슘 농도에 의해 유도되지 않습니다.
TRPC6 유전자의 결함은 Focal Segmental Glomerulosclerosis 2 (FSGS2)의 원인이 됩니다. TRPC6은 TRP(superfamily) 중 canonical TRP(TRPC) 계열에 속하며, PLC-coupled receptor에 의해 활성화되는 수용체 조절 양이온 채널의 구성 요소로 제안되었습니다.
TRPC6/6/7 하위군은 DAG 유사체에 의해 활성화될 수 있으며, PLC-coupled receptor를 통한 활성화 메커니즘을 시사합니다. Transfected cell에서 TRPC6은 비선택적 store-independent receptor-activated cation channel로 작용합니다. DAG에 의해 PKC-independent 방식으로 활성화되며 IP3 자극에는 반응하지 않습니다.
TRPC6은 생체 내에서 DAG-activated receptor-operated cation channel을 코딩하는 내인성 단백질로 여겨집니다. 관련 질환으로는 Glomerulosclerosis, Focal Segmental, 2 및 Focal Segmental Glomerulosclerosis가 있습니다.
