Product Specific Information
The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells, and is upregulated upon T-cell stimulation. CD27 binds to CD70 and plays an important role in T cell–B cell interaction.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for flow cytometry of normal human peripheral blood cells. Recommended at 5 µL (0.25 µg) per test (100 µL final volume).
Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.
Staining Panels:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Compensation:
Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads (Product # 01-2222-42) compared to single-color stained cells. In such cases, set compensation using cells. Similar observations with AbC Total Antibody Compensation beads (Product # A10497).
Light Sensitivity: Protect vial and samples from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information
CD27 is a 50 kDa member of the TNF receptor superfamily, including CD40 and CD30, which regulate cell proliferation and death. CD27 and its ligand CD70 are mainly expressed on lymphocytes. CD27 forms a disulfide-linked homodimer on mature thymocytes, peripheral T cells, and some B cells.
Activation of T cells via TCR-CD3 complex upregulates CD27 expression and releases soluble CD27 (sCD27), a prognostic marker for B cell malignancies. CD27-binding protein (SIVA) mediates apoptosis through CD27. Diseases associated with CD27 dysfunction include Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
