Product Specific Information

Description:
UCHL1 monoclonal antibody recognizes human CD45RO, a 180 kDa isoform of CD45. CD45RO is expressed by most thymocytes, activated memory T cells, granulocytes, and monocytes. CD22 acts as a ligand for CD45RO. Expression of CD45RO and CD45RA is used to distinguish peripheral T cell subsets.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (1.0 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Compensation Guidance:
Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads (Product # 01-2222-42). Prefer compensation using cells. Similar results observed with AbC Total Antibody Compensation beads (Product # A10497).

Light Sensitivity:
Protect vial and stained samples from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD45RO (UCHL1, ubiquitin carboxyl terminal esterase L1) is the shortest isoform of CD45, a receptor-type protein tyrosine phosphatase. CD45 regulates lymphocyte development and antigen signaling, modulates Src-family kinases, promotes cell survival, and participates in apoptosis-related DNA fragmentation. Isoforms vary by extracellular domain and expression depends on cell type and physiological state. CD45RO is expressed on macrophages, CD8⁺ T cells, activated T cells, and myeloma cells.
