Product Specific Information

Description:
The 1C10 monoclonal antibody reacts with mouse CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype switching and preventing apoptosis in combination with IL-4.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Use ≤1 µg per test (100 µL final volume). Optimize empirically for cell number (10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Use a 780/60 bandpass filter. Ensure instrument compatibility.

Compensation Guidance:
Compensation values for Super Bright 780-conjugated antibodies may differ between UltraComp eBeads microspheres and stained cells. For accurate results, set compensation with cells. Similar observations apply to AbC Total Antibody Compensation Beads.

Staining Panel Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal signal loss.

Excitation/Emission:
405 nm / 780 nm (Violet Laser)

Target Information

CD40 is a single-chain glycoprotein belonging to the TNF receptor superfamily. It is expressed on all B cells except plasma cells, and also on epithelial cells, carcinomas, and dendritic cells. CD40 plays a crucial role in immune regulation, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. Dysregulation is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency. CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease pathogenesis.
