The R1-2 monoclonal antibody reacts with mouse CD49d, the 150 kDa integrin alpha4 subunit. CD49d forms a non-covalent complex with integrin beta1 (CD29), known as VLA-4, which serves as a receptor for fibronectin and VCAM-1 (CD106). It is expressed by thymocytes, peripheral lymphocytes, monocytes, and eosinophils. CD49d also associates with integrin β7 and binds to MAdCAM-1.

• Tested by flow cytometry on mouse splenocytes

Target Information

Integrin alpha 4 (CD49d, ITGA4) is part of the integrin alpha chain family. Integrins are heterodimeric transmembrane proteins composed of alpha and beta chains. Alpha4 associates with either beta1 or beta7 chains, and the ligand-binding site resides on the alpha4 chain. Ligands include MadCAM, VCAM, and fibronectin. MadCAM is the primary ligand for integrin alpha4beta7. HIV-1 envelope has been shown to mimic MadCAM by binding and signaling through integrin alpha4beta7, the gut mucosal homing receptor for peripheral T cells.
