Description:
The monoclonal antibody miap301 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein with a small cytoplasmic domain. It is involved in adhesion through interactions with SIRP (signal regulator protein) and is associated with β3 integrins (CD51/CD61, CD41/CD61). CD47 also acts as a receptor for thrombospondin. T cell expression of CD47 can mediate activation or apoptosis in the presence of high levels of thrombospondin. Expression is found in most hematopoietic cells (T, B, monocytes, platelets, erythrocytes) and in non-hematopoietic cells.

No cross-reactivity is seen against human CD47.

Applications Reported:
This miap301 antibody has been reported for use in flow cytometric analysis and functional assays (blocking cell aggregation in vitro).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1 µg per 10⁶ cells in 100 µL total staining volume. Optimal titration is advised for best performance.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed cell surface transmembrane glycoprotein that interacts with several integrins to regulate their functions. Engagement of CD47 by ligands or counter receptors modulates signaling pathways, including activation of heterotrimeric G proteins and intracellular calcium increase upon cell adhesion. CD47 serves as a receptor for the C-terminal cell-binding domain of thrombospondin and may play roles in membrane transport and signal transduction.

CD47 has broad tissue distribution, with reduced expression on Rh erythrocytes. It acts as a ligand for CD172a (SIRPα), an inhibitory receptor on macrophages, preventing phagocytosis of CD47-positive cells. The CD47-CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, being abundant in synapse-rich brain and retinal regions.
