Description:
The 9F10 monoclonal antibody reacts with human CD49d, the 150 kDa integrin alpha4 subunit. The complex of CD49d with integrin beta1 (CD29), also known as VLA-4, acts as a receptor for fibronectin and VCAM-1 (CD106). This complex is expressed by thymocytes, peripheral lymphocytes, monocytes, and eosinophils. CD49d also associates with integrin beta7 and binds to Mucosal Addressin Cell-Adhesion Molecule-1 (MadCAM-1).

Applications Reported:
The 9F10 antibody has been reported for use in flow cytometric analysis and in vitro functional assays.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg/test (100 µL final volume). Optimal performance should be determined empirically.

Storage and Handling:
Use in a sterile environment.

0.2 µm post-manufacturing filtered.

Purity:

90%, as determined by SDS-PAGE.

Target Information

Integrin alpha 4 (CD49d, ITGA4) belongs to the integrin alpha chain family. Integrins are heterodimeric membrane proteins composed of alpha and beta chains. The alpha4 chain associates with either beta1 or beta7 chain, and the ligand-binding sites for both integrin alpha4beta1 and alpha4beta7 are located on the alpha4 chain. Ligands include MadCAM, VCAM, and fibronectin. MadCAM is the principal ligand for integrin alpha4beta7. HIV-1 envelope can mimic MadCAM by binding to and signaling through integrin alpha4beta7, the gut mucosal homing receptor for peripheral T cells.
