Product Specific Information

The monoclonal antibody B6H12 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein involved in adhesion via interactions with SIRP (signal regulatory protein) and integrins such as CD51/CD61 and CD41/CD61.

This interaction mediates bi-directional signaling, influencing neural synaptic activity and macrophage phagocytosis. CD47 acts as a receptor for thrombospondin, and its expression on T cells can mediate activation or apoptosis depending on thrombospondin levels. Stimulation of CD47 by monoclonal antibody induces CD4+CD25- suppressive activity and increases Foxp3 expression.

Expression is found in most hematopoietic cells (T, B, monocytes, platelets, erythrocytes) and in non-hematopoietic cells.
This antibody has neutralizing activity.

Applications Reported: Flow cytometric analysis and neutralizing assays.
Applications Tested: Tested by flow cytometric analysis of normal human peripheral blood cells, usable at ≤1 µg/test (in 100 µL final volume). Optimal titration recommended.

Storage and Handling: Use in sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).
Endotoxin Level: <0.001 ng/µg antibody (LAL assay).

Target Information

CD47 (Integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with integrins and regulates their functions. Engagement of CD47 modulates signaling pathways such as activation of heterotrimeric G proteins and intracellular calcium increase upon adhesion.

CD47 serves as a receptor for thrombospondin and plays roles in membrane transport and signal transduction. It is broadly distributed across tissues and reduced on Rh erythrocytes. CD47 binds CD172a (SIRP alpha), an inhibitory receptor on macrophages, preventing phagocytosis of CD47-positive cells.

The CD47–CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanical response modulation, and neuronal development, particularly in synapse-rich regions of brain and retina.
