Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon stress, p53 upregulates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 also relates to aging, oxidative stress, and cognitive functions such as long-term potentiation (LTP). It is expressed at low levels in normal cells but highly in transformed cell lines, contributing to malignancy. Mutations in TP53 cause loss of tumor suppressor activity and are linked to various human cancers, including Li-Fraumeni syndrome.
