Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 upregulates genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 consists of an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.
Phosphorylation at Ser15 is critical for activation and stabilization of p53. The protein plays a key role in preventing proliferation of damaged cells and functions as an anticancer agent. Phosphorylation at Ser392 has been linked to tumor formation. Elevated p53 levels are associated with aging, oxidative stress, and cognitive deficits due to altered long-term potentiation (LTP).
In normal cells, p53 levels are low, but in transformed or malignant cells, expression is high. Mutations in the TP53 gene can lead to loss of tumor suppressor activity and are common in human cancers. Germline mutations in TP53 are also found in cancer-prone families, such as those with Li-Fraumeni syndrome.
