Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for p53 activation and stabilization. p53 functions to block proliferation of damaged cells and acts as an anticancer agent. Phosphorylation at Ser392 is associated with tumor formation. p53 levels are low in normal cells but elevated in transformed cell lines, contributing to malignancy. Mutations in the TP53 gene are common in various cancers and lead to loss of tumor suppressor activity. Germline mutations can also occur in cancer-prone families such as those with Li-Fraumeni syndrome.
