Target Information
Smad4 단백질은 인간의 염색체 18번에 위치한 Smad4 유전자에 의해 암호화됩니다.
Smad4는 Smad family member 4, Mothers against decapentaplegic homolog 4 (MAD homolog 4)로도 알려져 있습니다.
이 단백질은 N-말단의 MH1 (MAD homology 1) 및 C-말단의 MH2 (MAD homology 2) 도메인을 포함하며, 각각 DNA 결합 및 단백질 상호작용에 관여합니다.
TGF-beta 및 BMP 수용체의 활성화는 R-Smad (Smad1, 2, 3, 5, 8)의 인산화를 유도하고, 이들이 Smad4 (co-Smad)와 삼합체를 형성하여 핵으로 이동합니다.
Smad4는 Cyclin D1의 발현 억제 및 collagen 발현 촉진을 통해 세포 증식을 억제하는 종양 억제 단백질로 기능합니다.
탈인산화는 Smad 단백질의 핵 내외 이동 및 동역학을 조절합니다.
