Target Information
Smad4 단백질은 인간 18번 염색체에 위치한 Smad4 유전자에 의해 발현됩니다.
Smad4는 Smad family member 4, Sma- and Mad-related protein 4, Mothers against decapentaplegic (MAD) homolog 4로도 알려져 있습니다.
이 단백질은 N-말단의 MH1 (MAD homology 1) 도메인과 C-말단의 MH2 (MAD homology 2) 도메인을 가지며, 각각 DNA 결합 및 단백질 상호작용에 관여합니다.
TGF-β 및 BMP와 같은 TGF-beta superfamily 리간드가 수용체에 결합하면, Smad1/2/3/5/8 (R-Smad)이 인산화되고 Smad4 (co-Smad)와 복합체를 형성해 핵으로 이동합니다.
Smad6 및 Smad7과 같은 억제성 Smad(I-Smad)는 활성화된 R-Smad와 상호작용하여 신호 경로를 조절합니다.
Smad4는 Cyclin D1 억제, Collagen 발현 증가 등을 통해 세포 증식을 억제하는 종양 억제 단백질로 작용합니다.
탈인산화 과정은 Smad 단백질의 핵 내·외 이동을 조절합니다.
