Target Information

The CDKN2A (p16INK4a) gene produces several transcript variants differing in their first exons. At least three alternatively spliced variants encode distinct proteins, two of which act as CDK4 kinase inhibitors. Another variant encodes an alternate open reading frame (ARF) that stabilizes the tumor suppressor protein p53 by interacting with and sequestering MDM2, an E3 ubiquitin-protein ligase responsible for p53 degradation.
Despite structural and functional differences, both CDK inhibitor isoforms and the ARF product contribute to cell cycle G1 control through regulation of CDK4 and p53.
Mutations or deletions in this gene are common in various tumors, marking it as a key tumor suppressor gene.
