Product Specific Information

The 17B5 monoclonal antibody reacts with mouse CD137 (4-1BB), a member of the TNFR superfamily. CD137 is expressed on activated T cells as monomers, dimers, and tetramers around day 4–6 after activation through the TCR complex. Interaction with its ligand, 4-1BBL, delivers a costimulatory signal crucial for T-APC interaction.
Recent data suggests CD137 is expressed on Tregs and may play a role in their proliferation. The 17B5 antibody exhibits strong antagonistic activity in functional assays.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested on stimulated mouse splenocytes; use ≤ 0.25 µg per test (100 µL final volume). Optimize antibody titration for best results.

Target Information

CD137 (TNFRSF9, 4-1BB) is an inducible costimulatory molecule mainly expressed on activated T cells. Its ligand, 4-1BBL, is found on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitors.
CD137 signaling maintains activated and memory CD8+ T cell survival, promotes clonal expansion, and regulates cytokine release affecting monocyte survival. Soluble CD137 may act as a negative regulator of immune responses.
