Product Specific Information

Description:
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 includes the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune regulation in mice. It is mainly expressed on activated T and B lymphocytes. J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells.

In vivo, J43 (intact or Fab) enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes similar to PD-1-deficient conditions.

• Tested on stimulated mouse splenocytes.
• Recommended ≤ 0.25 µg per test (100 µL final volume).
• Optimal antibody titration recommended for specific assays.

Target Information

PD-1 (PDCD-1) is an inhibitory receptor expressed on activated T cells, regulating immune responses by binding to ligands PD-L1 and PD-L2 on antigen-presenting cells. This interaction suppresses T-cell activation in the absence of danger signals such as LPS. PD-1-deficient mice exhibit hyperactive immune responses and autoimmune diseases. Despite its predicted molecular weight, PD-1 often migrates at higher molecular weights in SDS-PAGE.
