Target Information
Upon entry into mitosis, the cell’s microtubule (MT) network forms the mitotic spindle, enabling chromosome segregation. Kinetochores (KT) on centromeric chromatin are essential for proper chromosome attachment to spindle MTs.
The spindle and kinetochore complex (SKA complex), composed of SKA1, SKA2, and SKA3, is required for stable KT-MT interactions and timely anaphase onset.
Depletion of SKA1 or SKA2 by siRNA removes both proteins from the KT without affecting KT structure. Cells lacking the SKA complex exhibit prolonged metaphase-like delay, underscoring its role in metaphase plate maintenance and spindle checkpoint silencing.
