Product Specific Information

The CX5 monoclonal antibody reacts with mouse NKG2D, a lectin-like molecule expressed on NK cells. Mouse NKG2D binds to RAE-1 (α, β, γ, δ, ε) and H60 molecules, and can costimulate NK activation receptors through DAP12/DAP10 adaptor molecules. NKG2D is expressed by spleen and liver NK cells, NK1.1⁺ thymocytes, in vitro activated LAK cells, and a subset of splenic NKT cells.

• Suitable for use in flow cytometric analysis.

Applications Tested:

• Tested by flow cytometric analysis of mouse splenocytes.
• Recommended ≤0.125 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).
• Optimize antibody titration for best performance.

Fluorophore Information:

• PE-eFluor® 610 can be excited with 488–561 nm lasers and emits at 607 nm.
• Recommended filter: 610/20 band pass (equivalent to PE-Texas Red®).
• Light sensitive; protect from light.

Fixation:

• Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
• Clone-specific performance should be verified empirically.

Target Information

Natural killer (NK) cells mediate lysis of tumor and virus-infected cells without prior activation. They regulate humoral and cell-mediated immunity and express several calcium-dependent (C-type) lectins involved in NK cell function. The NKG2 gene encodes a type II transmembrane protein with a C-type lectin domain, located within the NK gene complex region containing multiple NK-specific lectin genes.
