Product Specific Information

Description:
The MAR-1 monoclonal antibody reacts with the Fc epsilon Receptor I alpha subunit (FcεRIα), an IgE-binding subunit lacking signal-transducing ability. FcεRIα is expressed on mast and basophil cells and is up-regulated by the presence of IgE. It forms a tetrameric complex with one beta and two gamma subunits containing immunoreceptor tyrosine-based activation motifs (ITAMs). The FcεRI complex is key in triggering IgE-mediated allergic reactions.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested in flow cytometric analysis of mouse bone marrow-derived mast cells. Recommended ≤0.25 µg per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Characteristics:
PE-eFluor® 610 is excited by 488–561 nm laser lines and emits at 607 nm. Recommended detection using a 610/20 band pass filter (equivalent to PE-Texas Red®). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

Excitation/Emission: 488–561 nm / 607 nm
Laser Compatibility: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered


Target Information

FceR1 (Fc epsilonR1 alpha) is a subunit of the high-affinity IgE receptor. It forms a tetrameric complex (αβγ₂), where the beta and gamma subunits mediate signal transduction. FcεRI plays a central role in allergic responses by coupling allergen-bound IgE with mast cells, leading to histamine and mediator release. This process initiates inflammatory responses characteristic of allergic disorders such as hay fever and asthma.
