Description:
The MIH12 monoclonal antibody reacts with human B7RP-1 (also known as B7h, B7-H2, GL50, ICOS Ligand). B7RP-1, a member of the B7 family, has a predicted molecular weight of approximately 40 kDa and belongs to the Ig superfamily. It is expressed by activated monocytes/macrophages and binds to the ICOS molecule expressed by activated T cells. The ICOS/B7RP-1 interaction is important in T cell costimulation.

Applications Reported:
This MIH12 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood monocyte-derived dendritic cells.
Use at 5 µL (0.125 µg) per test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test (empirically determined).

Fluorochrome Information:
PE-eFluor 610 can be excited with laser lines from 488–561 nm and emits at 607 nm.
Recommended filter: 610/20 band pass (equivalent to PE-Texas Red).
Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in:

Store up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

Inducible co-stimulator ligand (ICOSL) is a ligand on antigen-presenting cells and peripheral tissue that binds to the inducible co-stimulator receptor (ICOS).
ICOS belongs to the CD28/CD152 receptor family regulating T-cell activation and function.
ICOSL is expressed on monocytes, dendritic cells, and B cells, and can be induced by inflammatory stimuli.
Binding to ICOSL delivers a co-stimulatory signal promoting T cell proliferation and cytokine secretion.
