Target Information
The Ras p21 family of guanine nucleotide proteins plays a key role in signal transduction and is frequently mutated in human cancers. Ras proteins (H-, K-, and N-Ras p21) belong to a larger superfamily that includes Rap1A, Rap1B, Rap2, R-Ras, RalA, and RalB, which share about 50% amino acid homology with Ras. The six mammalian Rho proteins (RhoA, RhoB, RhoC, RhoG, Rho7, and Rho8) are approximately 30% homologous to Ras and are expressed in various cell types. Both Ras and Rho proteins contain a carboxy-terminal CAAX sequence (C = cysteine, A = aliphatic amino acid, X = any amino acid), which is essential for correct localization and function.
