Product Specific Information

Description:
The 2H7 monoclonal antibody reacts with human CD20, a 33–36 kDa transmembrane protein expressed by developing and mature B cells but not plasma cells. Low levels may be detected on a subset of mature T cells. CD20 plays a role in B-cell activation.

Applications Reported:
For use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (defined as staining a sample in 100 µL final volume).

Fluorophore Information:
PE-eFluor® 610 can be excited with laser lines from 488–561 nm and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488–561 nm / 607 nm / Blue, Green, Yellow-Green Laser.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 remains at the cell surface after antibody binding, making it a useful target for therapeutic B-cell depletion. Associated disease: Ms4a1-related common variable immune deficiency.
