Product Specific Information

Description:
The M1/69 monoclonal antibody reacts with the mouse CD24 molecule, also known as Heat Stable Antigen (HSA). This 35–50 kDa molecule is anchored in the plasma membrane via phosphatidylinositol and is expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage. CD24 is a variably glycosylated molecule resulting in heterogeneity of molecular mass on cells of different lineages. The expression of CD24 has been used to resolve stages of B lymphopoiesis in mouse bone marrow. It has been reported that P-selectin (CD62P) binds to CD24.

Applications Reported:
This M1/69 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended use ≤0.06 µg per test (100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Fluorochrome Information:
PE-eFluor 610 can be excited with laser lines from 488–561 nm and emits at 607 nm. Recommended filter: 610/20 band pass (equivalent to PE-Texas Red). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development, from pro-B to mature B-cells. Plasma cells show low or no expression. CD24 is also found in most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity control.
