Product Specific Information
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells, playing a key role in T cell development and activation. The GK1.5 binding is blocked by RM4-5.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; use ≤0.2 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment with NovaFluor dyes.
- This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which must be used for optimal staining.
- Add 5 µL per sample before combining with cells.
- For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
- NovaFluor conjugates use Phiton™ technology, providing engineered fluorescent signatures with minimal spillover and spread.

Target Information
CD4 is a co-receptor for MHC class II molecules, primarily expressed on T helper cells, and also found on monocytes, macrophages, and dendritic cells. It plays a central role in T-cell activation via association with the T-cell receptor complex and Lck kinase. CD4 expression is observed in thymus, lymph nodes, tonsils, spleen, and various non-lymphoid tissues. It may also mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been reported (RefSeq, July 2017).
