Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays an essential role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤0.2 µg/test).
A test is defined as the amount of antibody staining a cell sample in 100 µL volume. Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which contains formulation improvements for optimal staining.
Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.

Target Information

CD4 antigen is a co-receptor for MHC class II recognition and HIV binding. It is primarily expressed in T helper cells and also found in monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues such as brain and gut.
CD4 assists in early T-cell activation via association with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known.
