Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, a subset of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules and plays a critical role in T-cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck via its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤0.2 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA-intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD4 antigen is a co-receptor for MHC class II molecules and HIV. It is primarily expressed on T helper cells, but also on monocytes, macrophages, and dendritic cells. CD4 expression occurs in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. It aids in T-cell activation via interaction with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
