Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa surface receptor expressed by thymocytes, mature T cell subsets, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a crucial role in T-cell development and activation. In T cells, CD4 associates with tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤0.2 µg/test, 100 µL final volume, 10⁵–10⁸ cells/test)

Recommendations:

  • Carefully titrate antibody for optimal performance.
  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

  • Improved formulation over original CellBlox.
  • Required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample to antibody cocktails before cell addition.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence and minimal spillover.

Target Information

The CD4 antigen is a co-receptor for MHC class II recognition and HIV entry. It is expressed on T helper lymphocytes, monocytes, macrophages, and dendritic cells. CD4 is found in thymus, lymph nodes, tonsils, spleen, and specific regions of the brain and gut. It contributes to early T-cell activation via association with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
