Target Information
The tachykinins are a family of small peptides including the neurotransmitters substance P, neurokinin A, and neurokinin B. These peptides act on three distinct seven-transmembrane G-protein coupled receptors at different concentrations.
The NK-3 receptor (NK3R) has the highest affinity for neurokinin B and is highly expressed in the supraoptic and paraventricular nuclei. Upon ligand binding, NK3R activates a phosphatidylinositol-calcium second messenger system, leading to release of vasopressin and oxytocin.
NK3R may play a role in learning and memory, as mice lacking this gene show cognitive deficits. Although NK3R has been suggested to regulate vagal afferent relay neurons, it is more likely activated by substance P or neurokinin A, since airway nerves do not express neurokinin B.
