Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is breast ductal carcinoma.

Target Information

The progesterone receptor (PR) belongs to the steroid family of nuclear receptors and mediates the physiological effects of progesterone, which is crucial for reproductive events related to pregnancy establishment and maintenance.
PR exists as two isoforms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), derived from alternative translation initiation sites. PR-B is transcriptionally active and regulates genes responsible for endometrial maintenance, pregnancy, and ovulation inhibition.
PR-A differs from PR-B by a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity. In its inactive state, PR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, conformational change allows receptor dimerization and binding to progesterone response elements (PRE), leading to transcription activation.
A null mutation in the PGR gene can cause pleiotropic reproductive abnormalities.
