Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with the primary antibody for 30 minutes at room temperature. A suggested positive control is breast carcinoma tissue.
Target Information
The progesterone receptor (PR) belongs to the steroid family of nuclear receptors and mediates the physiological effects of progesterone, a key hormone in reproductive processes such as pregnancy establishment and maintenance.
PR exists as two isoforms: PR-A (94 kDa) and PR-B (120 kDa), generated by alternative translation initiation. PR-B is transcriptionally active and regulates genes involved in endometrial maintenance, pregnancy, and ovulation inhibition.
PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity. In its inactive state, PR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, PR undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription.
Mutations in the PGR gene can result in reproductive abnormalities.
