Product Specific Information

This antibody is predicted to react with bovine, chicken, human, mouse, non-human primate, Xenopus, rat, and zebrafish based on 100% sequence homology.
It is specific for the ~55 kDa Tryptophan Hydroxylase protein phosphorylated at Ser58 in Western blots of human dorsal Raphe nucleus.

Target Information

Tryptophan hydroxylase (TPH) catalyzes the 5-hydroxylation of tryptophan, the first step in the biosynthesis of indoleamines such as serotonin and melatonin.
In mammals:

  • Serotonin biosynthesis occurs predominantly in neurons originating in the Raphe nuclei of the brain.
  • Melatonin synthesis takes place within the pineal gland.

TPH activity is rate-limiting for serotonin but not for melatonin biosynthesis.
Serotonin acts mainly as a neurotransmitter, while melatonin is the principal hormone secreted by the pineal gland.
TPH activity is enhanced by phosphorylation via cAMP-dependent protein kinase (PKA) and Ca²⁺/calmodulin kinase II (CAM K II).
Both PKA and CAM K II phosphorylate Ser58 within the regulatory domain of TPH.
