- 인간 dorsal Raphe nucleus의 Western blot에서 Ser260에 인산화된 약 55 kDa Tryptophan Hydroxylase 단백질에 특이적입니다.
Target Information
Tryptophan hydroxylase (TPH)는 트립토판의 5-하이드록실화를 촉매하여 인돌아민(세로토닌 및 멜라토닌) 생합성의 첫 단계를 담당합니다.
포유류에서 세로토닌은 주로 뇌의 Raphe 핵에서 생성되고, 멜라토닌은 송과선에서 합성됩니다.
TPH는 두 조직 모두에서 동일한 반응을 촉매하지만, 세로토닌 생합성에서는 속도 제한 효소로 작용합니다.
TPH 활성은 PKA 및 Ca²⁺/Calmodulin kinase II (CAM K II)에 의해 인산화되어 증가하며, 두 효소 모두 TPH의 조절 도메인 내 Ser58을 인산화합니다.
