Product Specific Information

The AER-37 monoclonal antibody reacts with the Fc epsilon RI alpha subunit, an IgE-binding subunit lacking signal-transducing ability. FcεRI alpha is expressed on mast and basophil cells and is upregulated by the presence of IgE. It forms a tetrameric complex with one beta and two gamma subunits, which possess ITAM motifs responsible for signal transduction. The FcεRI complex plays a crucial role in triggering IgE-mediated allergic reactions.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry on peripheral blood cells at 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spectral spillover.

Target Information

FceR1 (Fc epsilonR1 alpha) is a subunit of the high-affinity IgE receptor. This tetrameric complex (α, β, and two γ subunits) is responsible for mediating allergic responses. When allergen-bound IgE molecules crosslink FcεRI receptors, mediators such as histamine are released, initiating inflammatory responses characteristic of allergic disorders like hay fever and asthma.
