Product Specific Information

Description:
The AER-37 monoclonal antibody reacts with the Fc epsilon RI alpha subunit, an IgE-binding subunit lacking signal-transducing ability. FcεRI alpha is expressed on mast and basophil cells and is upregulated by the presence of IgE. It forms a tetrameric complex with one beta and two gamma subunits that contain ITAM motifs, playing a key role in IgE-mediated allergic reactions.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of peripheral blood cells. Recommended use: 5 µL (0.4 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA-intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before mixing with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Based on Phiton™ technology utilizing nucleic acid dye structures engineered for optimized fluorescent signatures and minimal spillover.

Target Information

FceR1 (Fc epsilonR1 alpha):
A subunit of the high-affinity IgE receptor complex, consisting of one alpha, one beta, and two gamma subunits. The receptor mediates allergic responses by coupling allergen-bound IgE to mast cells, triggering histamine and mediator release responsible for hypersensitivity symptoms such as hay fever and asthma.
