Product Specific Information
The AER-37 monoclonal antibody reacts with the Fc epsilon RI alpha subunit, an IgE-binding subunit lacking signal-transducing ability. FcεRI alpha is expressed on mast and basophil cells and is upregulated by the presence of IgE. It forms a tetrameric complex with one beta and two gamma subunits, which contain immunoreceptor tyrosine-based activation motifs (ITAMs). The FcεRI complex plays a key role in triggering IgE-mediated allergic reactions.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of peripheral blood cells.
Use at 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
- Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining with NovaFluor conjugates.
- Add 5 µL per sample to antibody cocktails prior to combining with cells.
- For single-color controls, use 5 µL of buffer per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized fluorescent signatures and minimal spectral spillover.

Target Information
FceR1 (Fc epsilonR1 alpha) is a subunit of the high-affinity IgE receptor. The receptor is a tetrameric complex (one alpha, one beta, two gamma subunits). The beta and gamma subunits are essential for signal transduction. When allergen-bound IgE molecules crosslink multiple FcεRI receptors, mediators such as histamine are released, initiating allergic responses including hay fever and asthma. This process links allergens to mast cell activation and subsequent inflammatory responses.
