Heat-mediated antigen retrieval using a sodium citrate buffer (pH 6.0) is recommended for staining of paraffin sections. A suggested positive control for this product is breast carcinoma.
Mouse anti-human estrogen alpha antibody, clone 6F11, recognizes the human estrogen receptor alpha chain (ERα), also known as the estradiol receptor or nuclear receptor subfamily 3 group A member 1.
Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, upon binding estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding regulates gene transcription through coactivator or corepressor recruitment.
Two forms of estrogen receptor exist—alpha and beta—encoded by separate genes (ESR1 and ESR2). Alternative RNA splicing produces at least four ERα isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa).
ERs are expressed widely across tissues and are essential for sexual development, reproductive function, and bone physiology. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
