Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed on neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sialylated glycoproteins such as CD34, glycam-1, and MAdCam-1, mediating lymphocyte rolling and homing to peripheral lymphoid tissues.

• Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration recommended.

Fluorochrome Information:
Brilliant Violet™ 605 (BV605) emits at 605 nm and is excited by a 405 nm violet laser. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect the vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54). BV605 is not compatible with methanol-based fixation.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor composed of a lectin domain, EGF-like domain, SCR sequences, transmembrane, and cytoplasmic regions. It is constitutively expressed on most leukocytes including lymphocytes, monocytes, and polymorphonuclear cells, except for a subset of memory T cells.
