Product Specific Information

Description:
The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except mature erythrocytes and platelets. It plays a key role in lymphocyte activation via its tyrosine phosphatase enzymatic activity.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Validated by flow cytometric analysis of mouse bone marrow cells. Recommended usage ≤ 1.0 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Fluorochrome Information:
Brilliant Violet™ 605 (BV605) emits at 605 nm and is suitable for cytometers equipped with a 405 nm violet laser. Confirm instrument compatibility before use.

Buffer Recommendations:
When using multiple Brilliant Violet™, Brilliant Ultra Violet™, or Super Bright conjugates, use:

  • Super Bright Complete Staining Buffer (Product # SB-4401-42)
  • Brilliant Stain Buffer™ (Product # 00-4409-75)
    to minimize non-specific polymer interactions.

Light Sensitivity:
BV605 is photo-sensitive; protect vial and stained samples from light.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using:

  • IC Fixation Buffer (Product # 00-8222-49)
  • 1-Step Fix/Lyse Solution (Product # 00-5333-54)
    BV605 is not compatible with methanol-based fixation.

Target Information

CD45 (LCA, Leukocyte Common Antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, forming ~10% of lymphocyte surface proteins. Absent in non-hematopoietic tissues, CD45 regulates Src-family kinases in lymphocyte antigen signaling and influences T/B-cell receptor pathways. Multiple isoforms arise via alternative splicing with distinct extracellular domains. CD45 isoforms differ by cell type and physiological state, influencing membrane localization and signaling functions. It modulates integrin-mediated pathways, apoptosis-related DNA fragmentation, and various immune functions.
