Product Specific Information

This TULY56 monoclonal antibody reacts with human CD56 (Neural Cell Adhesion Molecule, NCAM). CD56 is a glycosylated transmembrane molecule expressed by neurons and NK/NKT cells, mediating homotypic adhesion of neural cells.
TULY56 does not block binding of CMSSB, suggesting different epitopes, and performs better after fixation and permeabilization. The antibody crossreacts with Rhesus macaque.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using human peripheral blood cells at 5 µL/test (0.25 µg). A test corresponds to staining a cell sample in 100 µL final volume (10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Violet™ 605 (BV605) emits at 605 nm and is designed for cytometers with a 405 nm violet laser. Protect from light due to photo-induced oxidation sensitivity.

Staining Buffer Recommendation:
When using multiple polymer-based dyes (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use:

  • Super Bright Complete Staining Buffer (Product # SB-4401-42) or
  • Brilliant Stain Buffer™ (Product # 00-4409-75)
    to minimize non-specific polymer interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

  • IC Fixation Buffer (Product # 00-8222-49), or
  • 1-step Fix/Lyse Solution (Product # 00-5333-54).
    BV605 is not compatible with methanol-based fixation.

Target Information

CD56 (NCAM) is a transmembrane glycoprotein of the immunoglobulin family expressed in the nervous system (120–180 kDa isoforms).
It mediates cell adhesion, migration, axonal growth, and synaptic plasticity.
CD56 is expressed on T cells, NK cells, and neuroectodermal-derived tissues and tumors (e.g., neuroblastoma, astrocytoma).
It serves as a neuroendocrine marker with high sensitivity for neuroendocrine and ovarian granulosa cell tumors.
Diseases associated with CD56 dysfunction include rabies and blastic plasmacytoid dendritic cell neoplasms.
